I am wondering if this setup will actually work

To be honest, I was looking at getting the
Wikipedia
but the warranty is only for 12 months, which is a bit thin 😢

Oh, I almost forgot. My desktop doesn't even have an eSATA port..

Oh right. My PC doesn't even have an eSATA port..

The setup will work fine. The enclosure uses SATA/USB, and the drive is SATA. 🙂
That larger second drive actually has a 36-month warranty—check the specs again. 😉
The only headache might be stock levels: if you head over to the Los Angeles-San Fernando Valley store, that first 500 GB drive is out of stock, and the 640 GB one is only available in San Francisco or Seattle. 🤔
